Microneedles are generally arranged in so-called microfibricated arrays. They are synthesized by reactive ion etching techniques on silicon or other types of material, in order to create microscopic arrays of needles, measuring 100-1000µm in length and 1-5µm in tip diameter. When these needles are inserted into skin, they pierce the stratum corneum and create microconduits for transport across the stratum corneum.

       Microneedles may be small enough to capture the convenience of patches but large enough to create micrometer-scale pathways across the skin for collagen delivery of even the largest macromolecules. Microneedles can be painlessly inserted into the skin of human subjects and their use does not need any medical expertise. The Dermaroller series share all of the advantages of the typical microneedle array, but have also a unique way of mechanically driving the applied substance under the skin, through the holes already created by the Dermaroller needles. The needles on the surface of the Dermaroller create miniscule holes of 0.07mm diameter and 0.13mm in depth, while the cylinder itself literally pushes mechanically the applied substance inside the holes and finally under the skin.

 

Macro-molecular delivery systems.

       On the first published study on the use of microfabricated microneedles to enhance product delivery across the skin. It is demonstrated that microneedles of 150µm length, arranged in dense arrays (150µm distance between microneedles) resulted in a 1,000-fold increase in the absorption of calcein by human skin, after being applied for only 10sec on cadaver human skin in one study. Removing the microneedles resulted in 10,000-fold increase, that is ten times more than leaving the microneedles in place, which means that microneedles have better results when the â€?â€?pierce and goâ€?â€? technique is used. (This is the actual technique used by the Dermaroller, which rolls over the skin, creating holes and pushing the substance inside the holes). This is how elastin fibers are recreated and collagen production is promoted.
